Bone Disease Modeling

Bone disease modeling is the use of experimental systems to reproduce the cellular, molecular, and structural features of skeletal disorders, enabling researchers to study disease mechanisms in controlled settings. Models may use cultured bone-forming osteoblasts, bone-resorbing osteoclasts, engineered tissues, or animals, with genetic, biochemical, or mechanical changes used to mimic abnormal bone remodeling. These approaches help connect disease-associated pathways to outcomes such as reduced bone formation, excessive resorption, or weakened tissue architecture. Bone disease modeling supports target identification, therapeutic testing, and evaluation of how biological and environmental factors influence skeletal health.

Animal models are frequently utilized to study cancer metastasis to bone. In this protocol we will describe two common methods of tumor inoculation for bone metastasis studies and briefly describe some of the analyses utilized to monitor and quantify these models.

Induction of Graft Versus Host Disease in Mouse Models via Allogeneic Bone Marrow and Splenic Cell Transplantation

2025

This video demonstrates the induction of intestinal graft-versus-host disease, GvHD, in a lethally-irradiated recipient mouse using allogeneic hematopoietic stem cells and GvHD-inducing splenic alloreactive T lymphocytes. This protocol helps establish a severe acute intestinal GvHD model to understand the mechanism of the disease and develop innovative therapeutic options.

Half-segmental Diaphyseal Bone Defect Model in Rats for Evaluating Bone Substitute Performance in Load-bearing Regions

2025

This study established a rat femoral half-segmental defect model to evaluate the mechanical and osteogenic performance of bone substitute materials under load-bearing conditions without the use of internal or external fixation.

Microhardness is a mechanical property and an informative parameter for evaluating hard tissue pathophysiology. Here, we demonstrate a standardized protocol (sample preparation, polishing, flat surface, and indentation sites) for microhardness analysis in tooth and alveolar bone in rodent oral disease models, namely, dental fluorosis, and ligature-induced periodontal bone resorption.

Generating Rabbit Model of Bone Infection: A Technique of Inducing Chronic Infection by Injecting Bacteria Directly in Bone Marrow

2025

This video describes the technique of surgically inducing bacterial infection in the bone marrow of a rabbit by using bone wax to avoid the leakage of bacteria. This rabbit model can be used to study bone infection treatment and bone regeneration.
