Bone Insufficiency Detection

Bone insufficiency detection is the identification of reduced bone strength or stress-related fractures that arise when normal skeletal tissue cannot withstand ordinary loading. It combines clinical assessment with imaging and, when appropriate, measurements of bone mineral density; radiography can reveal fracture lines, while magnetic resonance imaging detects early marrow changes and associated structural injury. In biology and medicine, these evaluations help distinguish insufficiency fractures from traumatic fractures, identify skeletal fragility before major injury, and guide decisions about monitoring, treatment, and fall or load management. Reliable detection also supports research on bone remodeling, aging, osteoporosis, and recovery after injury.

Automated Joint Space Detection Improves Bone Segmentation Accuracy

The development of an automated joint space detection workflow enabled high-throughput segmentation of distinct murine hindpaw bones with >98% accuracy in wild-type animals. Flexible application to forepaws and paws with inflammatory-erosive arthritis was achieved, but with deprecated performance that warrants further optimization in future studies using publicly available data.

Here we present a protocol for the dissection of hind limb long bones (femurs and tibiae) from the laboratory mouse. We further describe a rapid technique for bone marrow isolation from these bones that utilizes centrifugation for removal of bone marrow from the bone marrow space.
