Bone Marrow Transplants

Bone marrow transplants are medical procedures that replace damaged or diseased blood-forming tissue with healthy hematopoietic stem cells, helping rebuild the bone marrow and immune system. Before transplantation, patients often receive chemotherapy, radiation, or both to eliminate abnormal cells and suppress existing marrow, after which infused stem cells travel to the marrow and engraft, producing new red blood cells, platelets, and immune cells. Autologous transplants use the patient’s own cells, whereas allogeneic transplants use cells from a compatible donor and can provide an immune response against malignant cells. In biology and medicine, these procedures treat disorders such as leukemia, lymphoma, aplastic anemia, and certain inherited blood diseases, while also illustrating stem-cell renewal and immune regulation.

Bone marrow transplantation provides a way to change the genotype of the bone marrow derived cells. If the gene of interest is expressed in both bone marrow derived cells and non-bone marrow derived cells, bone marrow transplantation can change the bone marrow derived cells to a different genotype without changing the non-bone marrow derived cell genotype.

Bone marrow transplant is a potential cure for several diseases, including cancer and specific genetic disorders. Notably, this procedure is applicable for patients suffering from aplastic anemia, certain types of leukemia, severe combined immunodeficiency disease (SCID), Hodgkin's disease, non-Hodgkin's lymphoma, multiple myeloma, thalassemia, sickle-cell disease, and certain cancers. The transplant begins with high doses of chemotherapy and radiation treatment, which aim to destroy the...

This video describes the technique of aspirating bone marrow from iliac spine and smear preparation for biopsy. In the example, we perform bone marrow aspiration from an acute myeloid leukemia (AML) patient and examine the cells to check for measurable residual disease (MRD), an important prognostic biomarker.

Induction of Graft Versus Host Disease in Mouse Models via Allogeneic Bone Marrow and Splenic Cell Transplantation

This video demonstrates the induction of intestinal graft-versus-host disease, GvHD, in a lethally-irradiated recipient mouse using allogeneic hematopoietic stem cells and GvHD-inducing splenic alloreactive T lymphocytes. This protocol helps establish a severe acute intestinal GvHD model to understand the mechanism of the disease and develop innovative therapeutic options.
