C. Elegans Culture

C. elegans culture is the laboratory maintenance and propagation of the nematode Caenorhabditis elegans under controlled conditions, providing a tractable system for studying fundamental biology. Researchers sustain worms on prepared growth media with a bacterial food source, transfer populations between culture plates, and synchronize or collect animals at defined developmental stages for experiments. This approach supports investigations of development, genetics, aging, behavior, neurobiology, and disease-related pathways because C. elegans has a short life cycle, accessible anatomy, and powerful genetic tools. Reliable culture practices also improve reproducibility in gene-function studies, screening assays, and comparative research.

A Method for Culturing Embryonic C. elegans Cells

We describe here a revised protocol for large-scale culture of embryonic C. elegans cells. Embryonic C. elegans cells cultured in vitro using this method, appear to differentiate and recapitulate the expression of genes in a cell specific manner. Techniques that require direct access to the cells or isolation of specific cell types from the other tissues can be applied on C. elegans cultured cells.

The easiness of maintaining and propagating the nematode C. elegans make it a nice model organism to work with. The possibility of synchronizing worms allows the work with a significant amount of subjects at the same developmental stage, what facilitates the study of one particular process in many animals.

RNA interference (RNAi) is a widely used technique in which double stranded RNA is exogenously introduced into an organism, causing knockdown of a target gene. In the nematode, C. elegans, RNAi is particularly easy and effective because it can be delivered simply by feeding the worms bacteria that express double stranded RNA (dsRNA) that is complementary to a gene of interest. First, this video will introduce the concept of RNA interference and explain how it causes targeted gene knockdown.

To use Caenorhabditis elegans (C. elegans) in omics research, a method is needed to generate large populations of worms where a single sample can be measured across platforms for comparative analyses. Here, a method to culture C. elegans populations on large-scale culture plates (LSCPs) and to document population growth is presented.
