C. Elegans Embryonic Cells

C. elegans embryonic cells are the cells produced during early development of the nematode Caenorhabditis elegans, a tractable system for studying how a single fertilized egg forms a multicellular organism. Their development follows a largely invariant pattern in which asymmetric divisions, cell polarity, and cell-cell signaling establish distinct founder lineages and tissue fates. Researchers combine genetic manipulation, fluorescence microscopy, and time-lapse imaging to trace these cells and test the roles of developmental genes. Because embryonic divisions are rapid, reproducible, and optically accessible, C. elegans embryos provide a powerful model for investigating gene regulation, cell fate specification, morphogenesis, and conserved mechanisms of animal development.

We describe here a revised protocol for large-scale culture of embryonic C. elegans cells. Embryonic C. elegans cells cultured in vitro using this method, appear to differentiate and recapitulate the expression of genes in a cell specific manner. Techniques that require direct access to the cells or isolation of specific cell types from the other tissues can be applied on C. elegans cultured cells.

Here we detail protocols specifically designed to monitor morphogenic defects that occur during early and late phases of embryonic elongation of the nematode Caenorhabditis elegans. Ultimately, these protocols are designed to identify genes that regulate these phases and to characterize their differential requirements along the antero-posterior axis of the embryo.

The easiness of maintaining and propagating the nematode C. elegans make it a nice model organism to work with. The possibility of synchronizing worms allows the work with a significant amount of subjects at the same developmental stage, what facilitates the study of one particular process in many animals.

C. elegans Development and Reproduction

2023

Ceanorhabditis elegans is a powerful tool to help understand how organisms develop from a single cell into a vast interconnected array of functioning tissues. Early work in C. elegans traced the complete cell lineage and structure at the electron microscopy level, allowing researchers unprecedented insight into the connection between genes, development and disease. Appreciating the stereotyped development and reproductive program of C. elegans is essential to using this model organism to its...
