Calcium Activation

Calcium activation refers to a rise in intracellular calcium ions that acts as a signal to regulate cellular activity. In response to stimuli such as membrane depolarization or receptor activation, calcium enters through ion channels or is released from intracellular stores, then binds proteins such as calmodulin and triggers downstream enzymes, secretion, contraction, or gene expression. In biology, studying calcium activation helps explain muscle movement, neurotransmitter release, fertilization, immune responses, and cell growth. Fluorescent calcium indicators can track these transient signals, linking their timing and location to normal physiology and disease-related signaling changes.

This video showcases the assessment of B cell activation through the utilization of antigenic liposomes. B cells treated with calcium-sensitive Indo-1 dye are distinguished using flow cytometry, and their violet-to-blue fluorescence ratio is analyzed. As the cells encounter antigenic liposomes, it triggers B cell activation through calcium influx, progressively enhancing the violet-to-blue fluorescence signal ratio over time.
