Cardiomyocyte Specification

Cardiomyocyte specification is the developmental process through which cardiac progenitor cells acquire the identity and molecular program of heart muscle cells, providing the foundation for heart formation and function. During embryogenesis, coordinated signaling between pathways such as Wnt, BMP, and FGF guides mesodermal cells toward a cardiac fate, while transcription factors including NKX2-5, GATA4, and MEF2C activate cardiomyocyte-specific genes. Understanding this process supports the directed differentiation of pluripotent stem cells into cardiomyocytes for studying heart development, modeling inherited and acquired cardiac disease, screening potential therapies, and advancing regenerative medicine.

Isolation and Culture of Neonatal Mouse Cardiomyocytes

Primary mouse cardiomyocyte cultures are one of the pivotal tools for the investigation of myofibrillar organization and function. The following protocol describes the isolation and culture of primary cardiomyocytes from neonatal mouse hearts. The resulting cardiomyocyte cultures may be subsequently used for a variety of biomechanical, biochemical and cell-biological assays.

Measuring Fast Calcium Fluxes in Cardiomyocytes

We present a method to isolate rapid (microsecond) calcium events from slower fluxes in living cells using laser scanning confocal microscopy. The method measures fluorescence intensity fluctuations of calcium indicators by recording line scans of several hundred pixels in a cell. Histogram analysis allows us to isolate the time scales of different calcium fluxes.

The isolation of neonatal rat cardiomyocytes is a time consuming and unpredictable procedure. This study describes methods for cryopreservation and thawing of neonatal rat cardiomyocytes that allows for more efficient use of cells. The thawed NRCMs can be used for various experiments without the need for performing isolations each time.
