Carotid Artery Ligation

Carotid artery ligation is a surgical method that permanently or temporarily occludes a carotid artery, reducing blood flow to tissues supplied by that vessel and making it useful for studying vascular function. During the procedure, a suture or other ligature constricts the artery, lowering cerebral perfusion and creating changes in oxygen delivery, pressure, and collateral circulation. In biology research, this approach is used primarily in animal models to investigate cerebral ischemia, stroke-related injury, neurovascular responses, and tissue adaptation after altered blood supply. Measurements of behavior, blood flow, and pathology help connect vascular changes with cellular damage and recovery.

This describes a partial carotid ligation surgery, which causes disturbed flow conditions and subsequent atherosclerosis development (in two weeks) with intraplaque neo-vascularization (in four weeks) in the mouse common carotid artery. We also describe a novel method of RNA isolation from the carotid intima, providing high purity endothelial RNA.
