Cell Culture Media

Cell culture media are carefully formulated liquid environments that support the survival, growth, and specialized functions of cells outside the organism, making them essential to laboratory biology. They supply water, salts, nutrients, amino acids, vitamins, energy sources, and buffering components, while conditions such as pH, osmolarity, temperature, and gas exchange help maintain cellular homeostasis. Researchers select serum-containing or chemically defined media according to the cell type and experimental goal. Proper media design enables the expansion of primary cells and established cell lines for studying cell biology, disease mechanisms, drug responses, tissue engineering, and biotechnology, while reducing unwanted variation and improving experimental reproducibility.

Blaize1, Elizabeth Suter1 1 Department of Biological Sciences, Wagner College, 1 Campus Road, Staten Island NY, 10301 Prokaryotic cells are able to inhabit nearly every environment on this planet. As a kingdom, they possess a great metabolic diversity, allowing them to use a wide variety of molecules for energy generation (1). Therefore, when cultivating these organisms in the lab, all necessary and specific molecules required to make energy must be...

This video demonstrates a method for isolating and culturing microglia from rat brains. It entails separating microglia from other neuronal and non-neuronal cells using plates coated with a monoclonal antibody specific to microglia.

Exosome Isolation: A Density-based Ultracentrifugation Technique Using Sucrose Cushion to Separate Exosomes from Conditioned Media of Cultured Cells

2023

This video describes a density-based ultracentrifugation technique for isolating exosomes from conditioned media harvested from human embryonic kidney cell cultures using a 25% (w/w) sucrose cushion prepared in deuterium oxide. The isolated exosomes can be used for downstream applications.

The presence of stable microRNAs (miRNAs) in exosomes has generated immense interest as a novel mode of intercellular communication, for their potential utility as biomarkers and as a route for therapeutic intervention. Here we demonstrate exosome purification from blood and culture media followed by quantitative PCR to identify miRNAs being transported.
