Cell Culture Scale Up

Cell culture scale up is the controlled expansion of cells from small laboratory vessels to larger culture systems while preserving their growth, viability, and biological function. The process increases culture volume through sequential transfers or bioreactor expansion, while monitoring and adjusting cell density, nutrient availability, oxygen delivery, pH, temperature, mixing, and shear stress. Effective scale up maintains comparable culture conditions despite changes in vessel geometry and fluid dynamics. It supports the production of research materials, vaccines, therapeutic proteins, and cell-based therapies, and helps researchers evaluate how manufacturing conditions influence cell performance, product quality, reproducibility, and process efficiency.

Here, we present a robust, fast and scalable cardiomyocyte differentiation protocol for human pluripotent stem cells (hPSCs). Cardiomyocytes derived using this large-scale method can provide sufficient cell numbers for their effective use in human cardiovascular disease modeling, high-throughput drug screening, and potentially clinical applications.

This protocol describes the harvesting and visualization of elasmoid scales of zebrafish during in vivo regeneration. In addition, the ex vivo culture of these scales for up to 7 days after harvest is presented.

To use Caenorhabditis elegans (C. elegans) in omics research, a method is needed to generate large populations of worms where a single sample can be measured across platforms for comparative analyses. Here, a method to culture C. elegans populations on large-scale culture plates (LSCPs) and to document population growth is presented.
