Cell Isolation Protocol

A cell isolation protocol is a set of laboratory procedures used to separate viable cells from tissues, body fluids, or mixed cell populations for study or further culture. The process typically combines mechanical disruption with enzymatic digestion to break extracellular matrix and cell-cell connections, followed by filtration, centrifugation, washing, or selective methods that enrich the desired cells while limiting damage and contamination. In biology, isolated cells support microscopy, flow cytometry, molecular analysis, primary cell culture, and functional assays. Careful control of temperature, digestion time, and handling conditions helps preserve cell viability and identity, improving the reliability of downstream experiments.

Neutrophils are among the first cells to arrive on the site of inflammatory immune response, and their functions and mechanisms have been studied extensively in vitro. We demonstrate a standard density gradient separation method to isolate human neutrophils from whole blood using commercially available separation media.

Palatine tonsils are a rich source of B and T lymphocytes. Here we provide an easy, efficient and rapid protocol to isolate B and T lymphocytes from human palatine tonsils. The method described has been specifically adapted for studies of the viral etiology of tonsil inflammation known as tonsillitis.
