Cell Mapping

Cell mapping is the process of identifying, locating, and characterizing cells within a tissue, organism, or biological system, revealing how cellular organization supports function. Researchers combine microscopy, molecular markers, lineage tracing, and spatial transcriptomics to distinguish cell types and link their molecular profiles to precise positions and relationships. These maps can show how cells communicate, change during development, and respond to injury or disease. In biology, cell mapping supports studies of tissue architecture, developmental pathways, cancer biology, and neuroscience, while increasingly detailed cellular atlases provide reference frameworks for comparing healthy and diseased states.

A method is described to photoactivate single cells containing a caged fluorescent protein using two-photon absorption from a Ti:Sapphire femtosecond laser oscillator. To fate map the photoactivated cell, immunohistochemistry is used. This technique can be applied to any cell type.

Fate Mapping

Fate mapping is a technique used to understand how embryonic cells divide, differentiate, and migrate during development. In classic fate mapping experiments, cells in different areas of an embryo are labeled with a chemical dye and then tracked to determine which tissues or structures they form. Technological improvements now allow for individual cells to be marked and traced throughout embryonic development and adulthood. This video reviews the concepts behind fate mapping, and then details a...

We present a unique platform for characterizing electrode surfaces in solid oxide fuel cells (SOFCs) that allows simultaneous performance of multiple characterization techniques (e.g. in situ Raman spectroscopy and scanning probe microscopy alongside electrochemical measurements). Complementary information from these analyses may help to advance toward a more profound understanding of electrode reaction and degradation mechanisms, providing insights into rational design of better materials for...

Source: Laboratories of Jonas T. Kaplan and Sarah I. Gimbel—University of Southern California One principle of brain organization is the topographic mapping of information. Especially in sensory and motor cortices, adjacent regions of the brain tend to represent information from adjacent parts of the body, resulting in maps of the body expressed on the surface of the brain. The primary sensory and motor maps in the brain surround a prominent sulcus known as the central sulcus. The cortex...

Directed differentiation of hESCs into specific cells has generated much interest in regenerative medicine. We provide a concise, step-by-step protocol for determining the in vivo fate of selected hESCs that provides a valuable tool for characterizing tissue-specific reagents for cell-based therapy.
