Cell Proliferation Assay

Cell proliferation assays are laboratory methods used to measure how rapidly cells divide and increase in number, making them important for studying growth, development, disease, and treatment responses. Depending on the assay, proliferation is quantified by tracking cell number, DNA synthesis, or metabolic activity, often through a detectable colorimetric, fluorescent, or luminescent signal. Researchers use these assays to evaluate cell-cycle regulation, compare growth under different conditions, and test the effects of drugs, toxins, or genetic changes. Reliable proliferation measurements support cancer biology, drug discovery, tissue engineering, and other studies of cellular function.

The assay includes co-culturing MSCs with PBMCs to induce the monocyte population to acquire an immunomodulatory phenotype, leading to the secretion of interleukin-10 (IL-10). Following incubation with activated CD4+ T cells, the induced monocytes effectively suppress T cell proliferation.

Optimized Staining and Proliferation Modeling Methods for Cell Division Monitoring using Cell Tracking Dyes

Successful use of cell tracking dyes to monitor immune cell function and proliferation involves several critical steps. We describe methods for: 1) obtaining bright, uniform, reproducible label-ing with membrane dyes; 2) selecting fluorochromes and data acquisition conditions; and 3) choosing a model to quantify cell proliferation based on dye dilution.
