Chronic Lung Rejection

Chronic lung rejection is a progressive immune-mediated injury that can develop after lung transplantation, limiting long-term graft function and patient survival. The recipient’s immune system recognizes donor lung tissue as foreign, activating inflammatory cells and pathways that damage the small airways and other graft structures; repeated injury can produce airway narrowing, fibrosis, and persistent loss of airflow, often classified as chronic lung allograft dysfunction. Studying this process helps researchers understand transplant immunology, identify changes through lung-function monitoring and tissue analysis, and improve immunosuppressive strategies. Earlier recognition and better-targeted treatments may preserve graft performance and extend the benefits of transplantation.

Picrosirius Red staining is a semiquantitative method for objectively assessing collagen deposition in murine pulmonary fibrotic remodeling in a spatially resolved manner. Apart from evaluating total collagen deposition, Picrosirius Red staining allows for differentiating collagen fibers of different thicknesses.

Development of Obliterative Bronchiolitis in a Murine Model of Orthotopic Lung Transplantation

Obliterative bronchiolitis is the key impediment to the long-term survival of lung transplant recipients and the lack of a robust preclinical model precludes examining obliterative bronchiolitis immunopathogenesis. Unlike other solid organ transplants, vascularized mouse lung transplantation has only recently been developed. Here we show our independently developed obliterative bronchiolitis model after murine orthotopic single-lung transplantation.
