Clathrin Adaptor Interaction

Clathrin adaptor interaction is the coordinated binding between adaptor proteins, membrane lipids, cargo molecules, and clathrin that selects and concentrates materials for transport within eukaryotic cells. During clathrin-mediated endocytosis, adaptors such as AP complexes recognize sorting motifs on cargo and membrane phosphoinositides, while recruiting clathrin to assemble a coated pit that buds from the plasma membrane. These interactions regulate receptor internalization, vesicle formation, and intracellular trafficking between membrane compartments. Studying adaptor interactions helps explain how cells control nutrient uptake, signaling, and protein distribution, and provides a framework for investigating trafficking defects associated with disease.

Clathrin-mediated endocytosis, a rapid and highly dynamic process internalizes many proteins, including signaling receptors. The protocol described here directly visualizes the kinetics of individual endocytic events. This is essential for understanding how core members of the endocytic machinery coordinate with each other, and how protein cargo influence this process.
