Cns Progenitor Culture

CNS progenitor culture is an in vitro method for isolating, maintaining, and studying neural progenitor cells from the central nervous system, where these cells can self-renew and generate neuronal or glial lineages. In controlled culture conditions, defined growth factors and extracellular matrix support progenitor survival and proliferation, while changes in signaling cues or culture conditions can promote differentiation into neurons, astrocytes, or oligodendrocytes. This system enables researchers to examine neural development, lineage commitment, and disease-related cellular responses, and to test potential drugs or regenerative strategies. By linking cell behavior to specific molecular and environmental cues, CNS progenitor culture supports mechanistic studies across developmental and translational biology.

A Multi-compartment CNS Neuron-glia Co-culture Microfluidic Platform

We developed a novel multi-compartment neuron co-culture microsystem platform for in vitro CNS axon-glia interaction research. The platform is capable of conducting up to six independent experiments in parallel and was fabricated using a newly developed macro/micro hybrid fabrication method.

Primary, human fetal brain-derived, multipotential progenitor cells proliferate in vitro while maintaining the capacity to differentiate into neurons and astrocytes. This work shows that neural progenitors can be induced to differentiate through stages of the oligodendrocytic lineage by conditioning with select growth factors.

This video demonstrates the isolation and cultivation of cerebellar neural progenitor cells from a mouse pup's cerebellum. Cerebellar granule neuron progenitors are isolated and purified through differential adhesion, effectively removing astrocytes. These cultured progenitors subsequently differentiate into cerebellar granule neurons.
