Colonoid Culture

Colonoid culture is an in vitro method for growing three-dimensional, miniature colon tissues from intestinal stem cells, providing a controlled model of epithelial biology and disease. Cells are embedded in an extracellular matrix and supplied with defined growth factors that support stem-cell maintenance, proliferation, and self-organization into crypt- and lumen-like structures resembling aspects of the native colon. Researchers use colonoids to study epithelial renewal, host–microbe interactions, inflammation, infection, cancer, and drug responses. Because cultures can be generated from healthy or patient-derived tissue, they help connect basic mechanisms with personalized disease modeling and therapeutic testing.

Establishment of Human Epithelial Enteroids and Colonoids from Whole Tissue and Biopsy

We describe a method to establish human enteroids from small intestinal crypts and colonoids from colon crypts collected from both surgical tissue and biopsies. In this methodological article, we present the culture modalities that are essential for the successful growth and maintenance of human enteroids and colonoids.

We describe a protocol detailing the isolation of murine colonic crypts for the development of 3-dimensional colonoids. The established colonoids can then be terminally differentiated to reflect the cellular composition of the host epithelium prior to receiving an inflammatory challenge or being directed to establish an epithelial monolayer.

This video demonstrates New culture, a method by which chick embryos are cultured outside the egg for up to 24 hr. This method enables one to study early development (primitive streak to 14 som.), a period corresponding to E7-9 in mouse. Applications of this technique include electroporation, in situ hybridization and immunohistochemistry.
