Single Cell Learning

Single-cell learning is the capacity of an individual cell to modify its responses based on previous exposure to environmental stimuli, challenging the view that learning requires a nervous system. Through biochemical signaling, feedback networks, and changes in gene expression, cells can retain short- or long-lasting physiological states that influence later decisions, such as movement, growth, or stress responses. Studying this process in microorganisms and other individual cells helps explain cellular memory, adaptation, and decision-making. It also informs research on the origins of cognition, microbial behavior, synthetic biology, and how populations emerge from the actions of independently responding cells.

Learning new stimulus-response associations engages a wide range of neural processes which are ultimately reflected in changing spike output of individual neurons. Here we describe a behavioral protocol allowing for the continuous registration of single-neuron activity while animals acquire, extinguish, and reacquire a conditioned response within a single experimental session.

Understanding the function of the vertebrate central nervous system requires recordings from many neurons because cortical function arises on the level of populations of neurons. Here we describe an optical method to record suprathreshold neural activity with single-cell and single-spike resolution, dithered random-access scanning. This method records somatic fluorescence calcium signals from up to 100 neurons with high temporal resolution. A maximum-likelihood algorithm deconvolves the...

Here, we present a combinatorial approach for classifying neuronal cell types prior to isolation and for the subsequent characterization of single-cell transcriptomes. This protocol optimizes the preparation of samples for successful RNA Sequencing (RNA-Seq) and describes a methodology designed specifically for the enhanced understanding of cellular diversity.
