Colorimetric Detection

Colorimetric detection is a method that identifies or measures a substance by observing a color change, making invisible biological reactions easier to assess. In biological assays, a target analyte or enzyme reacts with a chromogenic reagent or substrate to produce a colored compound; the color intensity can then be estimated visually or measured as absorbance with a spectrophotometer. This approach supports detection of proteins, nucleic acids, metabolites, and enzyme activity in samples such as cells, tissues, and biological fluids. Its simplicity, low cost, and compatibility with microplate formats make colorimetric detection valuable for teaching laboratories, clinical testing, environmental biology, and quantitative research.

Source: Francis, M. B., et al. Detecting Cortex Fragments During Bacterial Spore Germination. J. Vis. Exp. (2016).This video demonstrates a colorimetric method to detect cortex degradation during bacterial spore germination by measuring the release of reducing sugars over time. Using N-acetylglucosamine standards and time-point samples, the assay quantifies sugar concentration through a purple chromogenic reaction, indicating the extent of cortex hydrolysis.

ampliPHOX colorimetric detection technology is presented as an inexpensive alternative to fluorescence detection for microarrays. Based on photopolymerization, ampliPHOX produces solid polymer spots visible to the naked eye in just a few minutes. Results are then imaged and automatically interpreted with a simple yet powerful software package.

Colorimetric Assay for Quantification of Lactate in Test Samples

This video describes a colorimetric assay for lactate measurement in cell lysates by using the lactate dehydrogenase enzyme. This enzyme converts lactate into pyruvate, along with the conversion of NAD to NADH that reacts with tetrazolium dyes and gives a colored product that can be measured spectrophotometrically to quantitate the lactate.

This video demonstrates a colorimetric E. coli detection method, involving an E. coli biomarker activating a DNAzyme, leading to cleavage of RNA linkage and detachment of a DNAzyme-urease conjugate. A subsequent colorimetric assay detects the E. coli biomarker through ammonia-induced pH changes.
