Continuous Culture Selection

Continuous culture selection is a biological method that enriches microorganisms with desired traits by maintaining a growing population in a continuously renewed culture system. Fresh medium enters while an equal volume of cells and waste leaves, and researchers control conditions such as nutrient limitation, dilution rate, temperature, or oxygen availability; organisms that grow faster or tolerate the imposed stress persist, whereas others are washed out. This approach supports the study of microbial competition, adaptation, and evolution under defined environmental pressures. It is also used to select strains with improved growth, metabolic activity, stress resistance, or production of valuable biological compounds.

Batch vs Continuous Culture

2026

Fermentation is a foundational biotechnological process used to produce pharmaceuticals, biofuels, enzymes, and food additives. Among industrial strategies, batch and continuous fermentation are the two most widely applied. Although both rely on microbial conversion of substrates into desired products, they differ markedly in operation, productivity, and suitability for specific applications.Batch fermentation occurs in a closed system in which nutrient media and inoculum are added at the...

Blaize1, Elizabeth Suter1 1 Department of Biological Sciences, Wagner College, 1 Campus Road, Staten Island NY, 10301 Prokaryotic cells are able to inhabit nearly every environment on this planet. As a kingdom, they possess a great metabolic diversity, allowing them to use a wide variety of molecules for energy generation (1). Therefore, when cultivating these organisms in the lab, all necessary and specific molecules required to make energy must be...

Continuous Culture of Bacteria at Constant Density via Automated Growth Rate Estimation

Begin with a turbidostat, a device that maintains a constant cell density in culture. The device contains a vessel with a bacterial culture, which is continuously aerated by stirring and maintained at a constant temperature.A laser diode emits a light beam that is split into two paths. One path is directed to a reference sensor to monitor laser intensity fluctuations, while the other passes through the culture.Bacterial cells scatter and absorb light, decreasing the transmitted intensity, which...

Batch and Continuous Bioreactors

2023

Bioreactors are used to grow organisms in large volumes, thereby enabling the production of mass quantities of the target product. These reactors can be batch reactors, which contain all of the components needed for cell growth, or continuous reactors, which have inlet and outlet ports allowing for the addition of fresh growth media and the removal of cell waste. This video presents batch and continuous reactors and demonstrates the use of bioreactors to grow bacteria in the laboratory.

Selectively Removing Microglia from a Rat Cerebellar Granule Neuron Culture

2025

This video demonstrates a technique for the selective removal of microglia from a primary rat neuronal culture. Cerebellar granule neurons are treated with an antimetabolite to eliminate proliferating glial cells. Additionally, they are treated with lysosome-targeting agents to deplete microglia from the culture.
