Copulatory Plug

A copulatory plug is a temporary mass of coagulated seminal fluid and reproductive secretions that forms in the female reproductive tract after mating, where it can influence fertilization and reproductive success. It develops when seminal proteins and other ejaculate components solidify within the tract, creating a physical barrier that may retain sperm, limit subsequent mating, or alter sperm competition. In biology, copulatory plugs are studied in rodents and other animals to investigate mating systems, sexual selection, sperm transport, and postcopulatory competition. Their presence can also help researchers identify recent mating and analyze how reproductive anatomy and behavior shape offspring production.

Bilayer Microfluidic Device for Combinatorial Plug Production

The fabrication of a polydimethylsiloxane (PDMS)-based bilayer device for the production of combinatorial libraries in water-in-oil emulsions (plugs) is presented here. The necessary hardware and software required to automate plug production are detailed in the protocol, and the production of a quantitative library of fluorescent plugs is also demonstrated.

The mucus plugged airways of cystic fibrosis (CF) patients are an ideal environment for microbial pathogens to thrive. The manuscript describes a novel method for studying the CF lung microbiome in an environment that mimics where they cause disease and how alterations of chemical conditions can drive microbial dynamics.
