Culture Preparation

Culture preparation is the process of establishing biological samples in controlled laboratory conditions so cells, tissues, microorganisms, or other living material can grow and be studied. It involves selecting an appropriate culture medium, preparing sterile vessels and equipment, introducing the sample, and maintaining conditions such as temperature, pH, nutrients, and gas exchange that support viability and growth. Aseptic technique helps prevent contamination that could alter culture behavior or compromise results. In biology, culture preparation supports cell and microbial research, disease modeling, biotechnology, drug testing, and the production of biological materials, providing reproducible systems for observing growth, responses, and interactions.

Chromosomes can be isolated from live cells such as lymphocytes or skin fibroblasts, and from organisms including humans or mice. These chromosome preparations can be further utilized for routine G-banding and molecular cytogenetic procedures such as fluorescence in situ hybridization (FISH), comparative genomic hybridization (CGH), and spectral karyotyping (SKY).

Source: Lin, P., et al. Synthesizing Sodium Tungstate and Sodium Molybdate Microcapsules via Bacterial Mineral Excretion. J. Vis. Exp. (2018).This video demonstrates the preparation of a monoclonal bacterial culture for mineral production through serial dilution and colony isolation on an agar plate. By shaking the plate with glass beads, individual cells are spatially separated to form genetically uniform colonies, which are then transferred to liquid media for growth. The culture with the...
