Cut And Tag

Cut and Tag is a chromatin-profiling technique that identifies the genomic locations of specific histone modifications or DNA-binding proteins using targeted sequencing. In permeabilized cells or nuclei, an antibody binds the protein or chromatin mark of interest, while a protein A or G-Tn5 transposase fusion attaches nearby and cuts accessible DNA, simultaneously adding sequencing adapters. The resulting fragments are purified and sequenced to map regulatory elements, nucleosome organization, and transcription-factor binding sites with high spatial resolution and low sample requirements. In biology, Cut and Tag supports epigenomic studies, cell-state characterization, developmental research, and investigations of gene regulation in limited or rare cell populations.

Researchers new to the epigenetic field will find CUT&Tag a significantly easier alternative to ChIP assays. CUT&Tag has tremendously benefited the epigenetic studies on rare and primary cell populations, generating high-quality data from very few cells.
