Cycloheximide Chase

Cycloheximide chase is a biochemical technique used to measure protein stability and estimate protein half-life in living cells or cell extracts. It first blocks new protein synthesis by inhibiting translation elongation, after which researchers collect samples at defined time points and track the decline of a target protein using methods such as immunoblotting. The resulting decay pattern reveals how quickly proteins are degraded and can indicate regulation by the ubiquitin-proteasome system, autophagy, or other turnover pathways. In biology, cycloheximide chase experiments help characterize protein regulation, signaling dynamics, and the effects of mutations, drugs, or cellular stress on protein stability.
