Death Inducing Signaling Complex

The death-inducing signaling complex (DISC) is a protein assembly that initiates extrinsic apoptosis, a regulated form of cell death triggered by signals outside the cell. When ligands such as Fas ligand or tumor necrosis factor bind cell-surface death receptors, the receptors cluster and recruit adaptor proteins, including FADD, through death-domain interactions. These adaptors assemble procaspase-8 and procaspase-10 at the DISC, promoting their activation and subsequent cleavage of downstream substrates that dismantle the cell. Studying DISC formation clarifies how tissues eliminate damaged or unwanted cells and supports research into immune regulation, cancer biology, and therapies that target apoptotic signaling.

This video demonstrates an ex vivo technique to induce brain cell death mediated through cytotoxic CD8+ T cells. Upon obtaining brain slices, activated oligodendrocyte-directed CD8+ T cells are transferred onto the slices to target the myelin sheath and oligodendrocytes.
