Decidua Tissue Peeling

Decidua tissue peeling is a specialized tissue-isolation technique that separates the maternal decidua, the pregnancy-associated uterine lining, from neighboring placental and fetal tissues for biological analysis. The procedure relies on identifying the boundary between maternal and fetal compartments and gently peeling or dissecting the decidual layer while minimizing contamination and preserving tissue architecture. Isolated decidua can then support studies of the maternal-fetal interface, including cellular composition, immune regulation, implantation, and placental development. By providing a more defined maternal tissue sample, this method improves investigations of pregnancy physiology and disorders such as abnormal placentation.

Described herein is a protocol to isolate and analyze the infiltrating leukocytes of tissues at the maternal-fetal interface (uterus, decidua, and placenta) of mice. This protocol maintains the integrity of most cell surface markers and yields enough viable cells for downstream applications including flow cytometry analysis.

This protocol presents two techniques to isolate the subcellular compartments of murine rod photoreceptors for protein analysis. The first method utilizes live retinae and cellulose filter paper to separate rod outer segments, while the second employs lyophilized retinae and adhesive tape to peel away rod inner and outer segment layers.
