Drosophila Indirect Flight Muscle

Drosophila indirect flight muscle is a specialized thoracic muscle system that powers flight and serves as a well-characterized model for studying muscle structure, function, and genetic regulation. Unlike direct flight muscles, these muscles do not attach to the wings; instead, their contractions deform the thorax, transmitting mechanical forces that move the wings, while stretch activation enables rapid, asynchronous contractions. Dissection of Drosophila melanogaster Indirect Flight Muscles for Microscopy Approaches

Drosophila is a powerful model to understand fundamental mechanisms of myogenesis. This protocol for the dissection and preparation of thorax hemi-sections enables microscopy analysis of indirect flight muscle from both pupal and adult stages. This protocol enables confocal imaging of cellular morphology, protein localization, muscle structure, and multiple other aspects of myogenesis.

We report a method to quantify muscle area, which is an indirect method to determine muscle mass in Drosophila adults. We demonstrate the application of our methodology by analyzing the indirect flight muscles in a Drosophila model of Myotonic Dystrophy disease.

Identification of mechanisms underlying muscle damage is crucial. Here we present the histological technique for preparing paraffin-embedded and frozen sections of Drosophila thoracic muscles. This allows analysis of muscle morphology and localization of protein and other muscle cell components.
