Drosophila Ovary Dissection

Drosophila ovary dissection is a laboratory technique used to isolate and examine the ovaries of the fruit fly, Drosophila melanogaster, a model organism for studying animal development and genetics. Under a stereomicroscope, researchers immobilize adult females, open the abdomen in physiological saline, and gently remove the paired ovaries while preserving their chain of developing egg chambers. The dissected tissue can then be fixed, stained, or cultured for microscopy and molecular analysis, allowing investigators to assess oogenesis, stem cell maintenance, cell differentiation, and tissue organization. This preparation supports research on reproductive biology, gene function, developmental signaling, and disease-related cellular processes.

The Drosophila ovary is an excellent model system for studying stem cell niche development. Though methods for dissecting larval and adult ovaries have been published, pupal ovary dissections require different techniques that have not been published in detail. Here we outline a protocol for dissecting, staining, and mounting pupal ovaries.

Drosophila oogenesis continues to be exceptionally useful in the study of mitochondrial proliferation and inheritance. This manuscript describes a detailed protocol used to label the replicating mitochondrial DNA (mtDNA) in Drosophila adult ovaries with 5-ethynyl-2´-deoxyuridine (EdU), which facilitates uncovering mechanisms associated with mitochondrial inheritance that were previously debatable.
