Drosophila Testes

Drosophila testes are paired reproductive organs in fruit flies that produce sperm and provide a tractable system for studying germline development, stem cells, and reproductive biology. Within the testis, germline stem cells divide asymmetrically to maintain a self-renewing stem cell and generate daughter cells that differentiate through spermatogenesis, while somatic support cells create a specialized microenvironment for this process. Because their cellular organization and genetic pathways are well characterized, Drosophila testes support research on stem cell maintenance, cell differentiation, meiosis, and tissue organization, offering insights into conserved mechanisms of development and fertility.

Drosophila melanogaster testes can be rapidly and efficiently isolated from adult males using dissecting needles. With practice, one can readily isolate in one or two days an amount of testes sufficient for the analysis of DNA or RNA by high throughput sequencing or more traditional molecular biology methods or of protein for antibody- or enzyme-based assays.

Drosophila Larval IHC

2023

Immunohistochemistry (IHC) is a technique used to visualize the presence and location of proteins within tissues. Drosophila larvae are particularly amenable to IHC because of the ease with which they can be processed for staining. Additionally, the larvae are transparent, meaning that some tissues can be visualized without the need for dissection. In IHC, proteins are ultimately detected with antibodies that specifically bind to “epitopes” within the protein of interest. In order to preserve...
