Ecm Stiffness

ECM stiffness is the resistance of the extracellular matrix to deformation, a physical property that helps regulate how cells organize, move, grow, and differentiate within tissues. Cells sense matrix rigidity through integrins and focal adhesions, which connect the ECM to the cytoskeleton and activate mechanotransduction pathways that alter cell shape, gene expression, and signaling. In biology and disease research, controlling or measuring ECM stiffness helps explain tissue development, wound healing, fibrosis, and tumor progression, while engineered matrices provide platforms for studying cell behavior and designing biomaterials.

An Assay to Study the Impact of Extracellular Matrix Stiffness on Bacterial Infection

2025

This video demonstrates an assay to study the effect of extracellular matrix (ECM) stiffness on bacterial infection of adherent cells. Polyacrylamide hydrogels of varying stiffness, overlaid with collagen, are layered in a multi-well plate to mimic the stiffness of physiological ECM. Upon overlaying human endothelial cells on the hydrogels and infecting the cells with Listeria monocytogenes bacteria engineered to express a fluorescent protein once intracellular, flow cytometry is used to...

Generation of Polyacrylamide and Silicone Extracellular Matrix Substrates with Defined Stiffness for Cell Biology Applications

2026

Although extracellular matrix (ECM) stiffness is a key regulator of cellular behavior, the underlying sensing and transduction mechanisms remain unclear. To address this, we provide protocols for creating polyacrylamide- and silicone-based substrates with tunable stiffness to advance our understanding of cell-ECM crosstalk.
