Egf Receptor Clustering

EGF receptor clustering is the spatial organization of epidermal growth factor receptors into groups within the plasma membrane, a process that influences how cells detect and respond to extracellular signals. When epidermal growth factor binds the receptor, it promotes receptor conformational changes, dimerization, and assembly into higher-order clusters that facilitate tyrosine phosphorylation and downstream signaling. These signaling platforms can regulate cell proliferation, survival, migration, and receptor internalization. Studying EGFR clustering with fluorescence imaging, biochemical assays, and quantitative modeling helps clarify cell communication and provides insight into abnormal signaling associated with cancer and other diseases.

This protocol describes the labeling of epidermal growth factor receptor (EGFR) on COS7 fibroblast cells, and subsequent correlative light- and electron microscopy of whole cells in hydrated state. The label contained fluorescent quantum dots. The protocol can be used to study the stoichiometry of EGFR at the single molecule level.
