Embryonic Cell Culture

Embryonic cell culture is the laboratory maintenance and growth of cells isolated from early embryos under controlled conditions, providing a model for studying development and cell behavior. Researchers place these cells on a suitable surface and supply nutrient-rich medium, growth factors, and regulated temperature, pH, and gas levels; these conditions support survival and proliferation while signaling pathways influence self-renewal or differentiation into specialized cell types. Embryonic cell culture enables investigation of gene regulation, tissue formation, and developmental processes, and supports stem cell research, disease modeling, and evaluation of potential therapies. Careful control of culture conditions improves reproducibility and helps link cellular changes to biological outcomes.

Here, we describe a method for isolation, culture and manipulation of mouse embryonic pancreas. This represents an excellent ex vivo system for studying various aspects of pancreatic development, including morphogenesis, differentiation and growth. Pancreatic bud explants can be cultured for several days and used in a range of different applications, including whole-mount immunofluorescence and live imaging.

Embryonic Stem Cell Culture and Differentiation

2023

Culturing embryonic stem (ES) cells requires conditions that maintain these cells in an undifferentiated state to preserve their capacity for self-renewal and pluripotency. Stem cell biologists are continuously optimizing methods to improve the efficiency of ES cell culture, and are simultaneously trying to direct the differentiation of ES cells into specific cell types that could be used in regenerative medicine. This video describes the basic principles of ES cell culture, and demonstrates a...

We describe here a revised protocol for large-scale culture of embryonic C. elegans cells. Embryonic C. elegans cells cultured in vitro using this method, appear to differentiate and recapitulate the expression of genes in a cell specific manner. Techniques that require direct access to the cells or isolation of specific cell types from the other tissues can be applied on C. elegans cultured cells.
