Eosin Methylene Blue

Eosin methylene blue is a combination of acidic eosin and basic methylene blue dyes used to differentiate biological materials and microorganisms. The dyes bind cellular structures according to their charge, producing contrasting colors, while in eosin methylene blue agar they also create a selective and differential environment: methylene blue and eosin inhibit many Gram-positive bacteria, and lactose fermentation lowers pH, causing characteristic dark colonies or a metallic green sheen in strong fermenters. This medium supports the detection and comparison of Gram-negative enteric bacteria in microbiology, including analysis of mixed cultures, environmental samples, and potential fecal contamination.
