Episome Integration

Episome integration is the incorporation of an episome, a DNA molecule that can replicate independently in a host cell, into the cell’s chromosomal genome. This process generally occurs through homologous or nonhomologous recombination, which joins episomal DNA with a chromosome and converts transient genetic material into a heritable genomic insertion. In biology, integration can alter gene expression, support stable transmission of introduced genes, or contribute to host genome changes during viral infection. Studying this process helps researchers understand genome stability, mobile genetic elements, gene regulation, and the design of persistent gene-delivery systems.

This protocol describes a detailed method for efficient generation of integration-free iPSCs from human adult peripheral blood cells. With the use of four oriP/EBNA-based episomal vectors to express the reprogramming factors, KLF4, MYC, BCL-XL, or OCT4 and SOX2, thousands of iPSC colonies can be obtained from 1 mL of peripheral blood.
