Ethanol Incubation

Ethanol incubation is a laboratory process in which biological samples, cells, or tissues are exposed to ethanol for a defined time and concentration to alter or preserve their chemical and structural properties. Ethanol removes water, disrupts lipid membranes, and denatures proteins, producing effects that depend on concentration, temperature, exposure time, and sample type. In biology, this treatment supports sample fixation, dehydration, permeabilization, preservation, and preparation for downstream staining or molecular analysis. Careful control of incubation conditions helps maintain reproducible morphology and biomolecular access while limiting excessive damage to the specimen.

A method to determine the time course of ethanol concentration in the brains of rats during operant ethanol self-administration is described. Gas chromatography with flame ionization detection is used to quantify ethanol in the dialysate samples, because it has the sensitivity required for the small volumes that are generated.

Biofuels: Producing Ethanol from Cellulosic Material

2023

Source: Laboratories of Margaret Workman and Kimberly Frye - Depaul University In this experiment, cellulosic material (such as corn stalks, leaves, grasses, etc.) will be used as a feedstock for the production of ethanol. The cellulosic material is first pretreated (ground and heated), digested with enzymes, and then fermented with yeast. Ethanol production is monitored using an ethanol probe. The experiment can be extended to optimize ethanol production by varying the feedstock used,...

Ethanol Resistance Assay to Evaluate Biofilm Matrix Integrity

Source: Bucher, T., et al. Methodologies for Studying B. subtilis Biofilms as a Model for Characterizing Small Molecule Biofilm Inhibitors. J. Vis. Exp. (2016).This video demonstrates how biofilm matrix integrity influences bacterial resistance to ethanol by comparing colony-forming units in untreated and inhibitor-disrupted biofilms. Reduced CFUs in inhibitor-treated samples demonstrate that the extracellular matrix is essential for protecting biofilm-embedded cells from antimicrobial stress.

Laboratory soil warming experiments usually employ two or more constant temperatures in multiple chambers. By presenting a sophisticated environmental chamber, we provide an accurate temperature control method to imitate the magnitude and amplitude of in situ soil temperature and improve the experimental design of soil incubation studies.
