Evans Blue Staining

Evans Blue Staining is a biological technique that uses a blue dye to visualize and measure vascular permeability in tissues. After administration, Evans blue binds strongly to serum albumin and normally remains within blood vessels; when endothelial barriers become compromised, the dye–albumin complex leaks into surrounding tissue, producing visible blue coloration that can be quantified by imaging or spectrophotometry. Researchers use this method to assess blood–brain barrier disruption, inflammation, edema, and tissue injury in experimental models. Because dye accumulation reflects macromolecular leakage, Evans blue staining provides a practical indicator of changes in vascular integrity and barrier function.

In this study, we describe a straightforward method to perform Evans Blue Dye (EBD) analysis on zebrafish larvae. This technique is a powerful tool for the characterization of skeletal muscle integrity and delineation of zebrafish models of muscular dystrophy, and is a valuable method for the development of novel therapeutics.
