Fluorescence Focus Assay

The fluorescence focus assay is a virological method that measures infectious virus by detecting groups of infected cells, or fluorescent foci, in cultured cell monolayers. After virus inoculation, infected cells produce viral proteins that are identified with virus-specific antibodies linked to fluorescent tags, allowing each focus to be visualized and counted as an indicator of infectious units. Researchers use the assay to quantify viral titers, evaluate antiviral compounds, compare viral infectivity, and assess neutralizing antibody activity. Its sensitivity and relatively rapid readout make it valuable for studying virus replication and host-pathogen interactions.

Low-Intensity Pulsed Ultrasound Stimulation (LIPUS) is a modality for non-invasive mechanical stimulation of endogenous or engineered cells with high spatial and temporal resolution. This article describes how to implement LIPUS to an epi-fluorescence microscope and how to minimize acoustic impedance mismatch along the ultrasound path to prevent unwanted mechanical artefacts.

The video demonstrates a procedure for quantifying the Zika virus in multiple mouse organs via a focus-forming assay. The process involves infecting mammalian cell monolayers with virus-infected tissue homogenates, followed by the use of methyl cellulose to prevent viral spread. Subsequently, immunostaining is employed to enable precise quantification and measurement of organ-specific infectivity.

(2022)This video demonstrates a focused LED-based assay for studying phototaxis in filamentous cyanobacteria. The bacteria migrate toward the light source, forming a dense accumulation at the illuminated center, which is visualized and quantified through pixel intensity analysis of the captured image.
