Fluorescently Labeled Markers

Fluorescently labeled markers are molecules or biological probes tagged with fluorescent compounds to make specific cells, proteins, nucleic acids, or structures visible, enabling researchers to track biological events. When illuminated with an appropriate excitation wavelength, the fluorophore absorbs energy and emits light at a longer wavelength, while the marker’s binding or targeting component identifies the feature of interest. Researchers use these markers in fluorescence microscopy, flow cytometry, immunofluorescence, and live-cell imaging to measure location, abundance, movement, and molecular interactions. These tools support studies of cell organization, gene expression, disease mechanisms, and responses to experimental treatments.

Here we describe a basic protocol for fluorescent labeling of different elements of heart tubes from larva and adult Drosophila melanogaster. These specimens are well-suited for imaging via fluorescent or confocal microscopy. This technique permits detailed structural analysis of the features of the hearts from a powerful model organism.
