Gef Gap Gdi

GEFs, GAPs, and GDIs are regulatory proteins that control the activity, localization, and signaling of small GTPases, molecular switches involved in processes such as cell growth, movement, and vesicle trafficking. GEFs activate GTPases by promoting GDP release and GTP binding, whereas GAPs accelerate GTP hydrolysis to return them to an inactive GDP-bound state; GDIs bind inactive, often lipid-modified GTPases and regulate their solubility and membrane cycling. Together, these regulators create precise spatial and temporal control of intracellular signaling. Studying the GEF-GAP-GDI system helps explain cytoskeletal organization, membrane transport, and how disrupted GTPase regulation contributes to diseases including cancer.

Affinity Precipitation of Active Rho-GEFs Using a GST-tagged Mutant Rho Protein (GST-RhoA(G17A)) from Epithelial Cell Lysates

The method presented here describes an assay to follow activation of RhoA specific GDP/GTP Exchange Factors (GEFs) in cultured cells by making use of a mutant RhoA GST fusion protein that has high affinity for activated GEFs. GEFs are precipitated from cell lysates, detected by Western blotting and quantified by densitometry.

Here, we describe the application of three-dimensional fluorescence recovery after photobleaching (3D-FRAP) for the analysis of the gap junction-dependent shuttling of miRNA. In contrast to commonly applied methods, 3D-FRAP allows for the quantification of the intercellular transfer of small RNAs in real time, with high spatio-temporal resolution.
