Gel Cassette Disassembly

Gel cassette disassembly is the controlled removal of a polyacrylamide or agarose gel from the plates, frames, or clamps that hold it during electrophoresis. After the run, the cassette is opened by releasing its seals or separating the plates, allowing the gel to be lifted without tearing, contamination, or loss of sample lanes. In biology, this step prepares gels for staining, imaging, band excision, protein transfer, or DNA recovery. Careful disassembly helps preserve band resolution and gel integrity, supporting accurate analysis in molecular biology, biochemistry, and genetic research.

Pouring and Running a Protein Gel by reusing Commercial Cassettes

Our protocol demonstrates how to pour multiple protein gels at a time by recycling Invitrogen Nupage Novex minigel cassettes, and inexpensive materials purchased at a home improvement store. This economical and streamlined method includes a way to store the gels at 4°C for a few weeks. By re-using the plastic gel cassettes from commercially available gels, labs that run frequent protein gels can save significant costs and help the environment.

Disassembly of influenza A virus cores during virus entry into host cells is a multistep process. We describe an in vitro method to analyze the early stages of viral uncoating. In this approach, velocity gradient centrifugation is used to biochemically dissect the steps that initiate uncoating under defined conditions.

Detection of Biofilm Disassembly Through a Dispersion Assay
