Glial Cell Culture

Glial cell culture is a laboratory method for growing and maintaining glial cells outside the nervous system, enabling controlled study of their biology and interactions with neurons. Researchers isolate or derive cells such as astrocytes, oligodendrocytes, or microglia, then provide suitable substrates, nutrients, and environmental conditions that support survival, proliferation, differentiation, or specialized functions. These cultures help investigate nervous system development, neuroinflammation, myelination, cellular signaling, and responses to injury or disease. By allowing researchers to manipulate defined conditions and measure cellular outcomes, glial cell culture supports studies of neural mechanisms, disease modeling, and evaluation of potential therapeutic strategies.

Obtaining a Mixed Glial Cell Culture from a Neonatal Mouse Brain

2025

The video demonstrated the method of isolating and culturing cells from a neonatal mouse brain. It involved enzymatic and mechanical dissociation of the brain, followed by culturing in growth media lacking neuronal growth factor to obtain a mixed glial population.

Establishing a Three-Dimensional Culture of Rat Brain-Derived Glial Cells

2025

This video demonstrates a technique to establish a three-dimensional culture of glial cells. Rat-brain-derived glial cells are encapsulated within a photocrosslinkable polymer matrix containing extracellular matrix components. Upon incubating in a growth medium, the glial cells proliferate within the supporting matrix to create a three-dimensional culture.

Establishing a Müller Glial Cell Culture Obtained from Mouse Retinas

2025

This video demonstrates the culturing of Müller glial (MG) cells isolated from mouse retinas. Retinas are dissociated into a single-cell suspension using digestive enzymes. Cells are then grown in a medium with a growth factor that promotes MG cell proliferation and neuronal cell mortality. Dead neuronal cells are removed while passaging, and MG cells are grown for further analysis.

Establishing Primary Mixed Glial Cell Cultures from a Mouse Spinal Cord

2025

This video demonstrates a technique for isolating and culturing primary mixed glial cells from adult mouse spinal cord tissue. The protocol involves enzymatic digestion of the tissue, followed by the removal of broken cell membranes and myelin debris to obtain a single-cell population. The cells are then cultured in media optimized for mixed glial cell growth.
