Gonadal Ridge Isolation

Gonadal ridge isolation is a microsurgical technique used to separate the embryonic gonadal ridges from surrounding tissues, enabling direct study of early gonad development. The procedure relies on careful dissection of the genital ridge, typically in association with the mesonephros, while preserving the tissue architecture and cell populations needed for analysis. Isolated ridges can be cultured, imaged, or processed for molecular and cellular assays to examine germ cell migration, somatic cell differentiation, and the formation of testes or ovaries. In developmental biology, this approach helps clarify the mechanisms of sex determination and provides experimental material for investigating reproductive disorders.

The ability of embryonic germinal cells to differentiate into primordial germinal cells during early development stages is a perfect model to address our hypothesis about cancer and infertility. This protocol shows how to isolate primordial germinal cells from developing gonads in 10.5-11.5 days post coitum mouse embryos.

Gonadal and Placental Hormones

2026

The gonads, namely the testes in males and the ovaries in females, are pivotal in producing gonadal hormones that orchestrate the intricate processes of sexual development and reproduction. In males, testosterone is the primary gonadal androgen. It plays a central role in the maturation of male reproductive organs — the penis and testes. Additionally, testosterone is instrumental in the development of secondary sexual characteristics — a deep voice as well as facial and pubic hair growth — and...
