Gst Tagged Proteins

GST-tagged proteins are recombinant proteins fused to glutathione S-transferase (GST), a soluble affinity tag that enables their detection, purification, and experimental handling. The GST domain binds specifically to immobilized glutathione, allowing fusion proteins to be captured from cell lysates on glutathione resin and subsequently eluted under mild conditions, often with free glutathione. This approach can also improve the solubility and stability of some expressed proteins, although the tag may alter their activity or interactions and sometimes requires enzymatic removal. In biology, GST-tagged proteins support protein purification, interaction studies, enzyme assays, structural research, and antibody production.

Affinity Precipitation of Active Rho-GEFs Using a GST-tagged Mutant Rho Protein (GST-RhoA(G17A)) from Epithelial Cell Lysates

The method presented here describes an assay to follow activation of RhoA specific GDP/GTP Exchange Factors (GEFs) in cultured cells by making use of a mutant RhoA GST fusion protein that has high affinity for activated GEFs. GEFs are precipitated from cell lysates, detected by Western blotting and quantified by densitometry.

In the present protocol, we demonstrate a highly efficient and cost-effective small-scale protein purification method, which allows purification of recombinant proteins by uniquely combining a cleavable GST-tag and a small His-tag.

We describe a method for the affinity-tagged purification of recombinant proteins using liquid-handling robotics. This method is generally applicable to the small-scale purification of soluble His-tagged proteins in a high-throughput format.
