Hev Nanoparticles

HEV nanoparticles are nanoscale assemblies derived from hepatitis E virus (HEV) structural proteins and are important tools for studying viral architecture, immunity, and vaccine design. They form when HEV capsid proteins self-assemble into ordered, virus-like particles that present viral surface features while lacking the viral genome required for replication. Because these particles can preserve key antigenic structures without producing infectious virus, researchers use them to investigate host recognition, antibody responses, and vaccine candidates. HEV nanoparticles also support structural biology and may inform the development of safer platforms for antigen delivery and antiviral research.

We have engineered the capsid protein of hepatitis E virus as a theranostic nanoparticle (HEVNP). HEVNP self-assembles into a stable icosahedral cage in mucosal delivery. Here, we describe the modification of HEVNPs for tumor targeting by mutating surface-exposed residues to cysteines, which conjugate synthetic ligands that specifically bind tumor cells.
