Human Bone Marrow Endothelial Cells

Human bone marrow endothelial cells are specialized cells that line the blood vessels of bone marrow and help organize the vascular niche that supports blood formation. Through selective barrier control, cell adhesion, and signaling interactions with hematopoietic and stromal cells, they regulate the movement, maintenance, and maturation of blood-forming cells within the marrow. Studying these cells helps clarify how vascular and hematopoietic systems interact in health and disease, including leukemia, bone marrow failure, and abnormal angiogenesis. Their isolation and culture also support research on drug responses, vascular biology, tissue engineering, and strategies for improving hematopoietic stem cell transplantation.

Isolating Mesangiogenic Progenitor Cells (MPCs) from Human Bone Marrow

Here we describe an optimized, highly reproducible protocol to isolate Mesodermal Progenitor Cells (MPCs) from human bone marrow (hBM). MPCs were characterized by flow cytometry and nestin expression. They showed the ability to give rise to exponentially growing MSC-like cell cultures while retaining their angiogenic potential.

Bioengineering of Humanized Bone Marrow Microenvironments in Mouse and Their Visualization by Live Imaging

A method to create and live-image different humanized bone-marrow niches in mice is presented. Based on the supportive niche created by human mesenchymal cells, the addition of human endothelial cells induces the formation of human vessels, while the addition of rhBMP-2 induces the formation of human-mouse chimeric mature bone tissue.
