Leaf Disc Homogenization

Leaf disc homogenization is a laboratory technique that mechanically disrupts circular pieces of leaf tissue to produce a uniform sample for biological analysis. During homogenization, cutting, grinding, or mixing breaks cell walls and membranes, allowing cellular contents to disperse throughout an extraction buffer and generating a representative tissue homogenate. In biology, this preparation supports the isolation or measurement of pigments, proteins, nucleic acids, enzymes, and other metabolites, helping researchers compare plant responses, investigate cellular composition, and prepare samples for downstream biochemical or molecular assays.

The goodness–of–fit test can be used to decide whether a population fits a given distribution, but it will not suffice to decide whether two populations follow the same unknown distribution. Unraveling how physiology and morphology are linked allows a deeper understanding of mechanistic functioning of plant leaves. We present both a procedure to derive parameters of stomatal regulation from stomatal conductance measurements and correlations with traditional functional leaf traits.

This manuscript describes how herbicide metabolism rates can be effectively quantified with excised leaves from a dicot weed, thereby reducing variability and removing any possible confounding effects of herbicide uptake or translocation typically observed in whole-plant assays.

The generation, purification and cell invasion of intracellular, cytoplasmic full length DISC1 protein aggresomes from cell cultures and of a labeled, multimeric recombinant DISC1 protein fragment in E. coli are described. Cell invasiveness is shown for recipient cells in cell culture and for neurons in vivo after stereotactical brain inoculation.
