In Vivo Two Photon Imaging

In vivo two-photon imaging is a fluorescence microscopy technique that visualizes cells and tissues within living organisms, enabling biological processes to be studied in their natural environment over time. It uses brief near-infrared laser pulses to trigger two-photon excitation only at the microscope’s focal point, producing fluorescence while limiting out-of-focus photodamage and improving imaging depth in scattering tissue. In biology, this approach supports live observation of neuronal activity, blood flow, immune-cell migration, and cellular interactions in intact tissues. Its capacity for repeated imaging links dynamic cellular behavior with tissue function and disease progression.

A minimally invasive protocol to stabilize the mouse spinal column and perform repetitive in vivo spinal cord imaging using two-photon microscopy is described. This method combines a spinal stabilization device and an anesthetic regimen to minimize respiratory-induced movements and produce raw imaging data that require no alignment or other post-processing.
