Inguinal White Fat

Inguinal white fat is a subcutaneous adipose tissue depot located near the groin and lower abdominal region, where it stores energy and contributes to whole-body metabolic regulation. Its white adipocytes accumulate triglycerides in large lipid droplets and release fatty acids during energy demand, while also secreting hormones and signaling molecules that influence insulin sensitivity, inflammation, and appetite. In rodents, inguinal white fat is a widely used model for studying adipose tissue remodeling because cold exposure or β-adrenergic stimulation can recruit beige adipocytes within the depot, increasing thermogenic activity. Research on this tissue supports investigations of obesity, diabetes, energy balance, and potential metabolic therapies.

Measurement of Basal and Forskolin-stimulated Lipolysis in Inguinal Adipose Fat Pads

This protocol describes the method of determining basal and forskolin-stimulated lipolysis in inguinal fat pads obtained from normal chow diet (NCD) or high fat diet (HFD) ± capsaicin fed wild type mice. As an index for lipolysis, glycerol release was measured from inguinal adipose fat pads.

Intravital Microscopy of the Inguinal Lymph Node

A technique for performing intravital microscopy of the inguinal lymph node (LN) is outlined. Such technique allows for real-time, in vivo study of the lymph node microvasculature and structure both during homeostasis and infection. This technique can be adapted to cell trafficking studies and to other lymph node sites.

Inguinal Subcutaneous White Adipose Tissue (ISWAT) Transplantation Model of Murine Islets

In this protocol, a method of murine islet isolation and transplantation into the inguinal subcutaneous white adipose tissue is described. Isolated syngeneic murine islets are transplanted into a murine recipient using a basement membrane hydrogel. The blood glucose level of the recipients is monitored, and histology analysis of the islet grafts is performed.

A Microphysiologic Platform for Human Fat: Sandwiched White Adipose Tissue

White adipose tissue (WAT) has critical deficiencies in its current primary culture models, hindering pharmacological development and metabolic studies. Here, we present a protocol to produce an adipose microphysiological system by sandwiching WAT between sheets of stromal cells. This construct provides a stable and adaptable platform for primary WAT culture.

Fat-Covered Islet Transplantation Using Epididymal White Adipose Tissue
