Interscapular Brown Fat

Interscapular brown fat is a specialized depot of brown adipose tissue located between the shoulder blades, where it generates heat and helps regulate body temperature. Cold exposure activates sympathetic nerves, increasing mitochondrial respiration in brown adipocytes; the mitochondrial protein uncoupling protein 1 (UCP1) dissipates the proton gradient as heat rather than driving ATP synthesis. This tissue is especially prominent in newborn mammals and small animals, whose limited insulation makes non-shivering thermogenesis essential. In biology research, interscapular brown fat provides a well-defined model for studying energy balance, mitochondrial function, temperature regulation, and the potential metabolic effects of activating brown adipose tissue.

A method of functional imaging of mouse brown adipose tissue (BAT) is described in which cold-stimulated uptake of 18F-Fluorodeoxyglucose (FDG) in BAT is non-invasively assessed with a standardized micro-PET/CT protocol. This method is robust and sensitive to detect differences in BAT activities in mouse models.

Rat Mammary Epithelial Cell Transplantation into the Interscapular White Fat Pad

2020

This article describes a transplantation method to graft donor rat mammary epithelial cells into the interscapular white fat pad of recipient animals. This method can be used to examine host and/or donor effects on mammary epithelium development and eliminates the need for pre-clearing, thereby extending the usefulness of this technique.
