Inner Segment Recording

Inner segment recording is an electrophysiological technique that measures membrane voltage or ionic currents from the inner segment of a sensory cell, most commonly a retinal photoreceptor. A recording micropipette forms a high-resistance seal with the cell membrane, allowing researchers to control membrane voltage or monitor current while the cell responds to light or other stimuli. This approach reveals how ion channels, membrane conductances, and signal-transduction pathways convert sensory inputs into electrical signals. In biology, inner segment recording supports studies of photoreceptor physiology, retinal circuitry, genetic disease mechanisms, and the effects of drugs or environmental conditions on cellular signaling.

Mesenteric afferent nerves convey information from the gastrointestinal tract towards the brain regarding normal homeostasis as well as pathophysiology. Gastrointestinal afferent nerve activity can be assessed by mounting isolated intestinal segments with attached afferent nerves into an organ bath, isolating the nerve, and assessing basal as well as stimulated activity.

Segmentation of three-dimensional data from many imaging techniques is a major bottleneck in analysis of complex biological systems. Here, we describe the use of SuRVoS Workbench to semi-automatically segment volumetric data at various length-scales using example datasets from cryo-electron tomography, cryo soft X-ray tomography, and phase contrast X-ray tomography techniques.
